I'm guessing it's a tooth problem.

We know that they can do fillings on ISS, so it would have to be way more serious than that, since per Administrator Isaacman, it was a "serious medical condition" (his words) during which it sounds like multiple people came to the crewman's assistance and ended with him being stable.
